Nous étudions les mécanismes moléculaires et cellulaires responsables des troubles cognitifs dans des maladies neurodéveloppementales (Déficience Intellectuelle, Coffin-Lowry, Dystrophies Musculaires) et dégénératives (Alzheimer). Notre approche multidisciplinaire chez des modèles murins est complétée par l’exploration préclinique de traitements induits par des modifications environnementales et des thérapies pharmacologiques et géniques.
Notre recherche est basée sur une approche intégrative, multi-échelle et translationnelle visant à :
(1) affiner notre compréhension des mécanismes moléculaires et cellulaires qui sous-tendent la plasticité cérébrale et les processus cognitifs,
(2) identifier les multiples déterminants et facteurs de risque (génétiques, développementaux, métaboliques) qui contribuent à leur détérioration et au dysfonctionnement des réseaux neuronaux dans des animaux modèles de maladies neurodéveloppementales (Déficience Intellectuelle, Syndrome de Coffin-Lowry, Dystrophies Musculaire de Duchenne) et neurodégénératives (Maladie d’Alzheimer),
(3) explorer le potentiel de stratégies de thérapie génique, pharmacologiques et comportementales offrant des perspectives réalistes. Ce programme de recherche, qui s’appuie sur notre expertise des mécanismes de plasticité et de cognition, a pour but de disséquer l’organisation fonctionnelle cérébrale et les mécanismes neuronaux sous-jacents aux fonctions de haut niveau qui se détériorent dans ces pathologies (cognition, émotion, fonctions exécutives). Notre approche multidisciplinaire est basée sur des modèles murins classiques et/ou de nouveaux modèles ; elle englobe des études comportementales (fonctions motrices, émotionnelles, cognitives, sociales et exécutives), la caractérisation de la plasticité cérébrale (plasticité synaptique et structurale, neurogenèse adulte) et de multiples déterminants contribuant à ces maladies, tels que la facteurs neurogénétiques, développementaux, non neuronaux (gliaux, vasculaires) et métaboliques, ainsi que des méthodes d’imagerie cérébrale in vivo, biochimie et signalisation cellulaire, imagerie confocale, biologie moléculaire, neurochirurgie, thérapie génique, pharmacologie… Nos projets s’inscrivent dans un contexte international hautement collaboratif incluant des interactions avec des clinicien.
